Ths purpose of this study was to examine the pharmacological effect of nortriptyline. the secondary amine antidepressant, on noradrenergic system from the point of pharmacokinetic and pharmacodynamics. Twelve healthy male volunteers took single
dose of
nortriptyline(20mg, 50mg). Plasma nortriptyine and MHPG concentration were measured periodically in 0, 1, 3, 5, 8, 12, 24, 28, 32, 48 hours after then by HPLS method. Summary of results were as follows:
1) Plasma peak levels of nortriptyline were attained at about 5 hours after administration of the drug. Area under the plama concentration time curve(AUC) was 297.7¡¾61.3hr ng/ml(20mg, N=6), 678¡¾102.hr ng/ml(50mg, N=6) respectively, and showed
proportional relationship with oral dose.
2) Plasma MHPG level was significantly decreased and maintained that condition from one to 48 hours, Reduction of MHPG level was largest in 8-12hours(up to 50%).
3) MHPG decrease showed slight delay compared with each nortriptyline concentration.
